So, I want to explore the “Hier Block” generate options in GRC, to
create a Python hierarchical block.
What should I terminate the “edges” of the graph with, and can I pass
parameters into the resulting block somehow?

“pad source” and “pad sink”.

On the parameters question: Just add a parameters block(s) to your Hier
block. When you add an instance of the Hier block to another FG, your
defined parameters will be available to modify from the outside.
